This depends on how you're using the tweet.  

If you're using information from an individual tweet, you'll need to cite it separately.  In MLA, the format is:

Lastname, Firstname (twitterusername). "The text of the entire tweet." year month, time of post. Tweet.

For example:

Library, MGHS (MGHSLibrary). "Our number 1 checkouts so far HAVE to be the graphic novels. #lovemygraphics". 20 Oct 2015, 14:53 UTC.

Using several individual tweets?  Use Tweet2Cite to generate your twitter citations.

If you want to cite an entire thread of tweets (perhaps an RSS feed, or a group of tweets found through a specific #hashtag search to gather a general understanding of views and ideas regarding a topic, you could cite the hashtag.
